石蠟制片法是將材料經(jīng)固定、脫水、透明、浸蠟后包埋在石蠟里面進行切片的方法。此法適用范圍，制片手段完備，能將材料切成極薄的片子（可切至2-8微米左右），并能制成連續(xù)的片子，這是其它制片法難以達到的，因此在光學顯微鏡的制片技術上它是zui常用的一種方法。除了一些經(jīng)不住石蠟切片法中所應用的各種藥劑處理的材料不能應用石蠟切片以外，一般的材料都可以用此法制片，但有制作時間較長，操作過程復雜以及材料易發(fā)硬或發(fā)脆等缺點。該法多采用旋轉式切片機進行切片。
石蠟制片操作程序如下：
（一）材料的采集與分割
采集材料應根據(jù)制片的目的和要求而定，材料要有代表性，無病蟲害或其它損傷，如要觀察病理解剖，則要取病斑、病癥部位。采下的材料應立即放在標本箱或濕布包起來帶回實驗室進行分割固定（如有條件也可在試驗地采集后進行分割固定）。為使固定液能迅速的滲透材料，材料要進行分割。分割時動作要迅速，以防材料萎蔫。分割塊的大小，宜小不宜大，一般不超過1cm3，分割后立即殺死固定。
