對照/標本無染色
① 確認是否忽略了應該加的某種試劑，包括一抗、二抗、三抗及底物等。
② 確認所有的試劑是否按正確的順序加入，是否孵育了足夠的時間。
③ 對照抗體的標簽確認是否使用了正確的抗體，以及所用的檢測系統(tǒng)是否和一抗匹配，這一點是非常重要的。比如，如果一抗是兔來源的抗體，二抗一定要用抗兔的二抗來匹配；或一抗是小鼠的IgM 一抗，二抗必須是山羊/兔抗小鼠的IgM（不是IgG）二抗。
④ 檢查抗體所使用的稀釋度及稀釋溶液。
⑤ 檢查抗體的有效期和保存條件，尤其是標記了酶或熒光素的抗體，現在大多數試劑公司的抗體均要求在4~8℃條件下保存，應避免反復凍融，試劑保存時一定要避免與揮發(fā)性有機溶劑同放一室，以免降低抗體的效價。
⑥ 檢查標本的儲存條件，用已知陽性的標本來同時做陽性對照。
⑦ 檢查色原/底物溶液，zui簡單的檢測方法是將一滴標記有酶的抗體加入到制備好的底物溶液中，如果底物發(fā)生預期的顏色變化，則可排除底物的因素。需要注意的是，有些底物在制成工作液后應在一定時間內用完，否則會失效。
⑧ 檢查沖洗液是否和反應試劑匹配，溶液的pH 值很重要，與過氧化物酶底物匹配的溶液中不應含有疊氮鈉。
⑨ 檢查復染劑和封片劑是否和所使用的色原匹配。

④ check the antibody used in the dilution and dilution solution.
⑤ check the validity of the antibody and storage conditions, especially labeled with enzyme or fluorescein antibody, and now most of the reagents companies require storage at 4 ~ 8 ℃ conditions, should be avoided repeated freezing and thawing, reagent preservation must be avoided And volatile organic solvents with a room, so as not to reduce the antibody titer.
⑥ check the specimen storage conditions, it is best to use known positive specimens to do positive control at the same time.
⑦ check the chromogen / substrate solution, the simplest detection method is to add a drop of enzyme labeled antibody added to the prepared substrate solution, if the substrate expected color change, you can rule out the substrate factor. It should be noted that some substrates should be used within a certain period of time after they are made into the working solution, otherwise they will fail.
⑧ Check whether the rinse solution and the reaction reagent match, the pH value of the solution is very important, and peroxidase substrate solution should not contain sodium azide.
⑨ check counterstain and packing agent and the color of the original match.
